By establishing a genetic rescue assay system in Drosophila, I showed for the first time that the enzymatic activity plays important roles in fine tuning transcriptional response in development. Loss of catalytic activity induces severe disruptions in embryonic segmentation gene expression, wing development, adult gene expression, and reduced viability, indicating an integration of the enzymatic activity in gene regulation.
A revision of my first-author manuscript describing this work is in review at XX journal. To further understand how the enzymatic activity may influence gene regulation at molecular level, I performed transcriptome analysis and identified genes specifically affected by the catalytic and the other mutants.
I am now following the preliminary data to study the role of E protein in regulating Drosophila innate immunity. Most importantly, I have gained the ability to perform and excel at independent research. The stem cell-like chromatin pattern of cancer cells and high frequency of epigenetic silencing of X target genes in human cancers ref suggest that X proteins may facilitate abnormal DNA hypermethylation.
I am very interested in studying how X proteins, together with Y, initiate DNA methylation to repress tumor suppressor genes. For example, do Ys progressively accumulate on X targets in embryonic carcinoma cells and cancer cells at genome-wide level?
Is DNA hypermethylation the only mechanism applied to silence X target genes? What is the molecular progression of this event? I believe my professional training in gene expression and technical skills, such as transcriptome analysis and chromatin immunoprecipitation ChIPprovide me with an excellent basis to carry out these functional studies.
